Item Germinação de sementes e estabelecimento in vitro de segmentos nodais de Libidibia Ferrea (Mart. ex Tul.) L. P. Queiroz(Universidade do Estado do Amazonas, 2023-03-17) Dantas, Alcineia dos Santos; Liberato, Maria Astrid Rocha; Silva, Daniel da; Batista, Ieda Hortencio; Ferreira, Francisca da SilvaThe species Libidibia ferrea (Mart. ex Tul.) L.P.Queiroz, popularly known as pau-ferro or jucá, belongs to the Fabaceae family and has great medicinal value due to the potential of its active principle, “pau-ferrol”. This species presents integumentary dormancy with difficulties in the production of seedlings, which impairs its propagation. Thus, the need arises to develop efficient protocols for the propagation of this important medicinal species. This work aimed to establish in vitro propagation methods by testing different substrates, different products for asepsis of explants, as well as the establishment of nodal segments of jucá in vitro. The research was carried out at the Laboratory of Tropical Silviculture and Digital Technologies (LASTED-INPA). Jucá seeds were provided by the Seed Bank of the Microbiology and Soil Fertility Laboratory of the National Institute for Research in the Amazon (INPA). The use of different substrates in the germination of L. ferrea seeds was evaluated, based on the analysis of the germination time period and the percentage of germinated seeds using two types of substrates. In addition, in vitro cultivation of Libidibia ferrea was carried out, using explants taken from germinated seedlings, testing different concentrations of sodium hypochlorite (NaCLO), 70% alcohol, Carbedazin® and Cabrio® Top fungicides. We observed that there were 53% of germinated seeds, testing the substrates organic soil and soil composed of organic with sand in the proportion 1:1, which favored the formation and development of uniform seedlings of L ferrea in ex vitro conditions. The results of the asepsis treatments with different chemical agents did not allow the establishment of a protocol. Keywords: jucá; tissue culture; medicinal plant; micropropagation
