Resumo

For the conservation of economically important microorganisms, biological collections have been shown to be quite efficient, because if conservation and maintenance techniques are adequate, the biological material will maintain its viable characteristics for prolonged periods. Therefore, the objective of this research was to reorganize a subcollection of the Microbiological Culture Collection Center of the Amazonas State University (CCM/UEA), seeking improvements in cell viability and improvement in the storage of preserved products. The development of the work was carried out in 4 stages, where 25 lineages preserved in Castellani and in mineral oil techniques were selected to carry out reactivation, identification at the gender level, maintenance and new preservation. Among the 25 lineages, 6 were unviable due to the error of the first conservations carried out previously or by the presence of contamination, making purification not possible, and 15 reached the conservation stage, but it was only possible to identify 9 lineages.
