dc.description.abstract | Dengue is an arboviral disease with a significant impact on global public health, particularly in tropical and subtropical regions such as Brazil. The high incidence of the disease, combined with the lack of specific antiviral treatments and the limited efficacy of available vaccines, underscores the need for new therapeutic strategies. In this context, the present study aimed to evaluate the antiviral activity of the peptide Glandicolin B, isolated from the fungus Penicillium rubens, in human hepatocytes (Huh-7) infected in vitro with dengue virus serotype 2 (DENV2). The research was conducted at the Cellular Immunopharmacology Laboratory (LIFCEL) of the State University of Amazonas. Initially, the cytotoxicity of Glandicolin B was assessed using the MTT colorimetric assay at various concentrations (0.1 to 100 µg/mL) and exposure times (24h, 48h, and 72h). The results indicated that the substance did not exhibit significant toxicity, maintaining cell viability above 90% at all tested concentrations. Subsequently, the Huh-7 cell line was infected with DENV-2 and treated with Glandicolin B. Antiviral activity was evaluated by quantifying the viral NS1 protein in the supernatants using the ELISA assay. The analysis showed a slight reduction in NS1 levels in treated cells, particularly at the concentration of 100 µg/mL, compared to the untreated infected group; however, the data did not reach statistical significance. Therefore, although Glandicolin B proved to be safe for use in Huh-7 cells, it did not significantly inhibit viral replication under the tested conditions. This result does not invalidate the potential of the compound but suggests the need for further experiments, including adjustments to incubation time, higher concentrations, coadministration with other compounds, or additional analyses involving other viral markers. dc.description.resumo | A dengue é uma arbovirose de grande impacto na saúde pública global, especialmente em regiões tropicais e subtropicais, como o Brasil. A elevada incidência da doença, aliada à ausência de tratamentos antivirais específicos e à limitada eficácia vacinal, reforça a necessidade de novas estratégias terapêuticas. Diante disso, este estudo teve como objetivo avaliar a atividade antiviral do peptídeo Glandicolina B, isolado do fungo Penicillium rubens, em hepatócitos humanos (Huh-7) infectados in vitro pelo sorotipo 2 do vírus da dengue (DENV2). O trabalho foi desenvolvido no Laboratório de Imunofarmacologia Celular (LIFCEL) da Universidade do Estado do Amazonas. Primeiramente, a citotoxicidade da Glandicolina B foi avaliada por meio do ensaio colorimétrico de MTT, em diferentes concentrações (0,1 a 100 µg/mL) e tempos de exposição (24h, 48h e 72h). Os resultados indicaram que a substância não apresentou toxicidade significativa, mantendo a viabilidade celular acima de 90% em todas as concentrações testadas. Em seguida, a linhagem Huh-7 foi infectada com o DENV-2 e tratada com a Glandicolina B. A atividade antiviral foi avaliada por meio da dosagem da proteína viral NS1 nos sobrenadantes, utilizando o teste de ELISA. A análise demonstrou uma discreta redução nos níveis de NS1 nas células tratadas, especialmente na concentração de 100 µg/mL, em comparação ao grupo infectado sem tratamento, mas os dados não apresentaram significância estatística. Assim, embora a Glandicolina B tenha se mostrado segura para uso em células Huh-7, não demonstrou inibição significativa da replicação viral sob as condições testadas. Este resultado não invalida o potencial da substância, mas sugere a necessidade de novos ensaios, com ajustes no tempo de incubação, concentrações mais elevadas, coadministração com outras substâncias ou análises complementares de outros marcadores virais.
